A Single Protein Block Rebuilt Worn Joints In Mice And Human Tissue As Stanford Drug Makes A Major Breakthrough

According to Stanford Medicine, scientists have regrown lost knee cartilage and prevented arthritis by blocking a single aging-related protein called 15-PGDH.

In older mice, the treatment rebuilt thinner, worn cartilage across the joint surface, and the regrown tissue was hyaline cartilage, the smooth type joints actually need. In a model mimicking ACL tears, mice treated twice weekly for four weeks were far less likely to develop osteoarthritis. Crucially, human cartilage taken during knee replacement surgeries also began producing new cartilage after one week.

This matters because osteoarthritis affects roughly one in five US adults and drives about $65 billion in healthcare costs yearly, yet no approved drug reverses it. An oral version is already in Phase 1 trials and proven safe for muscle weakness.

“The effect was remarkable,” said senior author Nidhi Bhutani.
