Scientists Map Biosynthetic Pathway Of Wolfsbane And Larkspur For First Time Opening Door To Lab-Made Versions Of Deadly But Medically Promising Compounds

Scientists Map Biosynthetic Pathway Of Wolfsbane And Larkspur: MSU And Czech Academy Team Identifies Six Key Enzymes That Build Diterpenoid Alkaloid Atisinium In Tobacco Biofactories. Aconitine Still Unsynthesised After 200 Years

According to EurekAlert / Michigan State University, researchers at Michigan State University and the Czech Academy of Sciences have for the first time mapped the biosynthetic pathway that wolfsbane and larkspur use to produce diterpenoid alkaloids; complex plant compounds that cause neurotoxicity and paralysis in tiny doses but hold potential against pain, malaria, cancer and pests. The findings are published in Molecular Plant.

Diterpenoid alkaloids are the intersection of the two oldest and largest classes of plant chemicals, and their structures are so complex that aconitine; isolated nearly 200 years ago, has never been successfully synthesised in a lab. The MSU-Czech collaboration used a molecular scavenger hunt through thousands of genes across multiple larkspur and wolfsbane species to identify which genes were active in the right tissues at the right time.

The team inserted a promising set of genes into tobacco plants, which acted as biofactories. Those plants produced a diterpenoid alkaloid called atisinium through six unique enzymes, revealing both the initial assembly steps and an unexpected nitrogen source.

“In an ideal scenario, this could eventually help create new drugs inspired by these natural products,” said Lana Mutabdžija of the Czech Academy. “Our vision is to provide green, sustainable tools that will allow us to harness these plants’ natural power,” added Professor Björn Hamberger of MSU.
